What Research Shows

Advanced paternal age is one of the most consistent environmental risk factors identified in autism research. A large meta-analysis found that fathers over 40 have a 28 percent higher risk of having an autistic child compared to fathers aged 25-29. The risk increases gradually with age rather than showing a sharp threshold. However, the absolute increase in risk is modest; the vast majority of children born to older fathers do not develop autism.

The Biological Mechanism

The primary explanation involves de novo mutations. Sperm cells divide continuously throughout a man’s life, and each division carries a small chance of new genetic mutations. By age 40, a man’s sperm has undergone significantly more divisions than at age 25, accumulating more mutations. Some of these mutations occur in genes relevant to brain development and have been linked to autism risk. A 2012 study found that the number of de novo mutations in offspring increases by approximately two per year of paternal age.

Putting Risk in Perspective

While the relative risk increase is statistically significant, the absolute risk remains low. If the baseline risk of autism is about 2.8 percent (1 in 36), a 28 percent increase raises it to roughly 3.6 percent. More than 96 percent of children born to older fathers will not be autistic. Paternal age is one of many factors in a complex genetic landscape, not a deterministic cause.

Maternal Age and Other Factors

Maternal age also shows some association with autism risk, though the relationship is weaker and more debated. Advanced parental age on either side may interact with other genetic and environmental factors. Fertility treatments, which are more common among older parents, have been studied as a potential confound but have not been identified as an independent risk factor.
